GT dPCR SARS-CoV-2 Variants of Concern Mutational Signature Assay Kit (Patent Pending)
200 Tests. Kit requires 3 reactions to be run for each sample. This assay contains all primers, probes, and controls for detection and discrimination of the signature mutations within the Spike protein-gene of the Alpha (UK, B.1.1.7), Beta (South African, B.1.351), Gamma (Brazilian/Japanese, P.2), Delta (Indian, B.1.617.2), and Epsilon (Californian, B.1.427/B.1.429) variants from other strains of the SARS-CoV-2 virus. While this test does not target every mutation present in these virus variants, it targets both parental- and variant- associated genomic sequences corresponding to Spike protein amino acids 69-70 and 501 (reaction 1), 417 and 484 (reaction 2), and 452 and 484 (reaction 3). The first reaction provides detection and discrimination of the signature mutations associated with the Alpha (N501Y and del69-70) variant from the parental (N501 and HV69-70) strain. The second reaction provides detection and discrimination of the signature mutations associated with the Beta (E484K and K417N) and Gamma (E484K and K417T) variants from the parental (E484 and K417) strain. The third reaction provides detection and discrimination of the signature mutations associated with the Delta (E484Q and L452R) and Epsilon (E484 and L452R) variants from the parental (E484 and L452) strain. Previous reports suggest these residues impact protein folding, and amino acid substitutions at these residues alter ACE-2 receptor binding and potentially promote immune system evasion. Kit includes validated GT-4-plex primer probe solution, GT-5-plex primer probe solution, GT-4-plex primer probe solution for Delta/Epsilon, plus ddPCR assay controls: GT-Parental (Wuhan) Control, GT-Alpha (UK) Control, GT-Beta (South African) Control, GT-Gamma (Brazilian/Japanese) Control, GT-Delta (Indian) Control, GT-Epsilon (Californian) Control, and the reaction-relevant GT- Variant Mixture Controls: Parental-Alpha-Beta-Gamma mixture for reactions 1 and 2, and Parental-Delta-Epsilon mixture for reaction 3.
